Induction of prophage SPO2 in Bacillus subtilis: prophage excision in the absence of bacterial or bacteriophage DNA synthesis.

Bacillus subtilis lysogenic for SPO2 wild type was induced under conditions preventing synthesis of both bacterial and phage DNA. The infectivity of phage DNA in transfection is strongly decreased under these conditions, whereas the activity of single phage genes as measured by marker rescue with superinfecting phage is unaffected. DNA from induced cells was sedimented in neutral sucrose gradie...

Bacillus subtilis bacteriophage SPbeta: localization of the prophage attachment site, and specialized transduction.

The attachment site for the prophage of SPbeta lies between ilvA and kauA on the chromosome of Bacillus subtilis strain 168. Specialized transduction of citK and kauA can be carried out by certain lysates of SPbeta.

Genetic mapping of a defective bacteriophage on the chromosome of Bacillus subtilis 168.

A genetic marker responsible for the killing activity of PBSX, a defective phage carried by Bacillus subtilis 168, has been located on the bacterial chromosome. Two mutant strains of B. subtilis 168, which produced tailless phage particles upon mitomycin C induction, were shown to carry lesions, designated xtl-1 and xtl-2, which were linked by transformation and PBS1-mediated transduction to me...

Congression of unlinked markers and genetic mapping in the transformation of Bacillus subtilis 168.

A thorough examination of cotransformation of two unlinked genetic markers in Bacillus subtilis 168 shows that the two recombinational events do not occur randomly. The cotransformation frequency is dependent on the distance between the two markers as well as on the order in which they replicate in the competent cell. These results indicate that uptake and/or integration of DNA molecules bearin...

Genetic mapping of a mutant defective in D,L-alanine racemase in Bacillus subtilis 168.

Genetic analysis of a d-alanine requiring mutant (dal) of Bacillus subtilis reveals that the gene that codes for d,l-alanine racemase is linked to purB. The order of genes in this region of the chromosome is purB, pig, tsi, dal. Thus there are at least two clusters of genes that regulate cell wall biosynthesis in B. subtilis.
